Anticancer and apoptosis-inducing activities of microbial metabolites Electron. J. Biotechnol.

The problems of systemic toxicity and drug resistance in cancer chemotherapy urge the continuing discovery of new anticancer agents. We explored the specific anticancer activity from microbial metabolites to find new lead compound. 394 microbial extracts were evaluated on anti-proliferative activity against 4 cancer cell lines using MTT assay. Of these, 20 samples showed varying degree of cytotoxicity but specifically to the cancer cell lines since the growth of normal cells was not significantly inhibited by 1 mg/ml of each cell extracts. The 4 most potent extracts exhibited strongest growth inhibition to each cancer cell type were selected for further studied. Abstract Background: Cancer is the second most common fatal disease in the world, behind cardiovascular disorders in the first place. It accounts for around 0.3 million deaths per year in India due to the lack of proper diagnostic facilities, prevention and treatment. Current therapeutic methods do not provide adequate protection and affect normal cells along with cancerous ones. Chemical profile, agaritine and selenium content of Agaricus bisporus BABT

Chemical profile of the Agaricus bisporus samples were analyzed using GC/MS method in ethanolic extracts. A total of 174 metabolic products were detected, which included 13 significant metabolites between 1.2 to 83 % (w/w); other 13 metabolites at 1% (w/w) and 148 metabolites less than 1% classified into 12 categories. These metabolites had many medicinal activities, which included anti-cancer, anti-cardiovascular diseases, anti-hypercholesterol, anti-microbial, hepatoprotective, human health supporting and immune enhancer. HPLC analysis of water extracts of the A. bisporus samples showed that the spores and gills in the fresh adult mushroom had higher percentage of agaritine mycotoxin than the other parts. ABSTRACT The synthesis of 3,3-disubstituted N-methyloxindoles, starting from 3-acetyl-2-hydroxy-1-methyloxindole employing a sequential one-pot synthesis, is studied. The process involves a first alkylation in the presence of 1 equiv. of both organic halide and Triton B and the second one employs another 1.5 equiv. of each in moderate to high yields. This procedure is compared with the results obtained from the direct dialkylation of N-methyloxindole. The metathesis of one of the corresponding diallylated product was also studied obtaining the spiranic oxindole. All these methodologies are directed towards the access to anticancer agents and natural biologically active products.

Screening of Metarhizium spp. strains for anticancer indolizidine alkaloid production and its rapid detection by MS analysis BJM

Six fungi strains (M. anisopliae 3935, 4516, 4819, PL57, PL43 and M. flavoviride CG291) were studied regarding their ability to produce an anticancer indolizidine alkaloid. The culture process was carried out in Shaken flask at 26ºC and 200 rpm using three different culture medium containing oat meal extract supplemented with glucose and DL-lysine or Czapek culture medium. The mycelial extracts produced by Metarhizium spp. cultures were directly submitted to electrospray ionization mass spectrometry (ESI-MS) analysis and the highest alkaloid concentration (approximately, 6 mg.L-1) was reached when M. anisopliae 3935 was tested.

Targeting of temozolomide using magnetic nanobeads: an in vitro study BJPS

Temozolomide, a chemotherapeutic drug that is often administered for the treatment of brain cancer has severe side effects and a poor aqueous solubility. In order to decrease the detrimental effect of the drug over healthy cells, a novel drug delivery vehicle was developed where the therapeutic drug was encapsulated within the hydrophobic cavities of b-CD modified magnetite nanoparticles, which are embedded in chitosan nanobeads prepared by salt addition. BACKGROUND: Novel, in silico-designed anticancer compounds were synthesized in our laboratory namely, 2-ethyl-3-O-sulphamoyl-estra-1,3,5(10),15-tetraen-17-ol (ESE-15-ol) and 2-ethyl-3-O-sulphamoyl-estra-1,3,5(10)16-tetraene (ESE-16). These compounds were designed to have improved bioavailability when compared to their source compound, 2-methoxyestradiol. This theoretically would be due to their increased binding affinity to carbonic anhydrase II, present in erythrocytes.
